12,13-DiHOME is a hydroxylated derivative of linoleic acid, characterized by two hydroxyl groups at the 12th and 13th carbon positions of the octadecenoic acid chain. The molecule features a cis configuration (Z) at the 9th double bond. Its molecular formula is C18H34O4, and its molecular weight is 314.46 g/mol. Due to its amphipathic nature, 12,13-DiHOME exhibits both hydrophobic and hydrophilic interactions, which play a crucial role in its biological activity. Elevated levels of 12,13-DiHOME are associated with certain pathological conditions, including acute respiratory distress syndrome (ARDS) and cardiovascular diseases. Its concentration in the blood often increases in response to tissue injury or stress, indicating its potential as a biomarker for oxidative stress and inflammation. Additionally, the modulation of 12,13-DiHOME levels is being explored as a therapeutic target for metabolic diseases, such as obesity and diabetes, where its effects on lipid metabolism and insulin sensitivity could provide beneficial outcomes.
The quantification of 12,13-DiHOME in biological samples is typically performed using mass spectrometry-based techniques, such as liquid chromatography-tandem mass spectrometry (LC-MS/MS). These methods offer high sensitivity and specificity, enabling the detection and quantification of 12,13-DiHOME at low concentrations. This analysis is crucial for understanding its physiological and pathological roles, monitoring its levels in various diseases, and evaluating the effects of potential therapeutic interventions.
