    4D-DIA Proteomics Analysis

      Traditional proteomics technologies such as iTRAQ, TMT, and Label-free are all based on DDA (Data Dependent Acquisition) mode. In DDA mode, typically only the top 10-20 parent ions of highest abundance in the first-level mass spectrometry are selected for fragmentation, and their secondary mass spectrometry is printed for qualitative analysis. Since the DDA mode does not collect information on all ions, some proteins with lower abundance are often ignored, which to some extent limits our comprehensive understanding of the proteome. In contrast, DIA (Data Independent Acquisition) mode detects and fragments all ions to ensure that all ion information in the sample is obtained without omission and without difference.

       

      4D-DIA proteomics is an emerging high-throughput mass spectrometry technology. It is based on the 4D-proteomics platform of timsTOF Pro2, introduces the dimension of ion mobility on the basis of DIA, not only significantly improves the accuracy and repeatability of quantitative analysis, but also achieves high stability and high precision quantitative analysis of proteomics in large sample queues.
