Absolute Quantification Analysis of Vitreous Humor Tissue Proteins

The vitreous humor is a gel-like transparent substance located at the back of the eye, which maintains the shape of the eye and plays a crucial role in preserving the health of the retina. The protein component in the vitreous humor can reflect the health status of the eye, including retinal disease, glaucoma, and ocular complications of systemic diseases like diabetes. Therefore, a detailed analysis of its protein composition is vitally important for ophthalmological research. The Absolute Quantitation of Proteins (AQUA) technique is a method for accurately measuring the absolute amount of specific proteins or peptides. By using synthetic, known concentration of stable isotope labeled peptides as internal standards, AQUA allows researchers to obtain accurate concentration information of proteins.

      Sample: Vitreous Humor Tissue

In ophthalmology research, the proteomic analysis of vitreous humor tissue is key to unraveling the mechanisms of retinal disease. Since vitreous extraction is an invasive procedure for patients, it is essential to ensure that each extracted sample is utilized to the max. The AQUA technology was born in this context, for the realization of efficient, precise protein analysis.

      Technology: Protein Absolute Quantitation Analysis

Absolute quantitation realizes absolute quantification by using synthetic peptides labeled with stable isotopes: based on the discovery of the relationship between mass spectrometric signal and protein concentration, the average mass spectrometric signal response of the three strongest trypsin peptides in each mole of protein is constant, with a coefficient of variation less than 10%. As long as an internal standard (synthetic peptide labeled with a question-and-answer isotope) is given, the relationship between mass spectrometric signal and protein concentration can be used to determine the universal signal response factor (counts/mol). The common signal response factor for all measured proteins is the same.

The convenience and sensitivity of this method, together with the widespread use of tandem mass spectrometers, make the AQUA absolute quantitative analysis strategy an effective method for directly determining protein levels from cell lysates.

      Service Advantage

      1. Accuracy

Due to the use of isotope-labeled internal standards, the AQUA technique can provide highly accurate quantitative information, avoiding the many variable factors in relative quantification.

      2. Sensitivity

This technique is particularly effective for detecting low-abundance proteins in the vitreous, giving it tremendous potential for early disease diagnosis and monitoring.

      3. Standardization

AQUA provides a standardized method for comparing protein expression between different samples, which is essential for clinical research and biomarker discovery.

      4. Reproducibility

The strategy of using an internal standard ensures consistent results, even under different experimental conditions.

      Application

      1. Research on the Molecular Mechanisms of Ophthalmic Diseases

      2. Evaluation and Monitoring of Drug Treatment Effects

      3. Development of Biomarkers for Retinal Diseases

      4. Formulation of Personalized Treatment Plans for Patients
