Dehydrolithocholic acid (DLCA) is an oxidized metabolite derived from lithocholic acid, primarily found in bile. In the liver, lithocholic acid undergoes enzyme-mediated oxidation to form DLCA, a crucial step in the bile acid metabolic pathway. As an intermediate in this pathway, DLCA plays a significant role in the synthesis, detoxification, and excretion of bile acids. It is also essential in maintaining the enterohepatic circulation of bile acids and ensuring normal liver function. Specifically, DLCA aids in preventing bile acid-induced cellular damage by promoting the excretion of hydrophobic bile acids and reducing their cytotoxic effects. Fluctuations in DLCA levels are closely linked to various hepatobiliary diseases and metabolic disorders, such as cholestasis, cirrhosis, and hepatobiliary cancers. Therefore, the precise quantification of DLCA is vital not only for understanding its role in bile acid metabolism but also for early diagnosis of liver diseases, monitoring disease progression, and conducting drug metabolism research.
