Eggshell Protein Glycosylation Sites and Sugar Type Analysis

When we boil an egg for breakfast every morning, few people ponder the scientific mysteries hidden behind this seemingly ordinary food. In fact, the glycosylation sites and sugar types within the eggshell provide us with a unique window to explore biology and food science.

      The Magic of Eggshell Proteins

An eggshell is not just a physical barrier, it is a complex biological structure composed of various proteins and sugar molecules. These proteins and sugar molecules interact with each other, determining the strength, flexibility, and permeability of the eggshell. The glycosylation sites play a crucial role in this process, enabling proteins to bind tightly with other molecules.

Glycosylation Sites and Their Importance in Eggs

Protein glycosylation is a common modification of proteins, which means adding one or more sugar molecules to specific amino acid residues. This addition can increase the stability, solubility, and affinity of proteins. In the proteins of eggshells, the existence of glycosylation sites endows them with specific functions and characteristics.

      Services at Mtoz Biolabs

In order to explore the glycosylation sites of eggshell proteins in depth, MtoZ Biolabs has led the application of a series of high-end technologies, including liquid chromatography-mass spectrometry. This technology can identify and quantify each glycosylation site and its related sugar type in the proteins of eggshells, providing unmatched accuracy and sensitivity.

Our experienced technicians usually choose multiple enzymes to digest the target protein in order to cover the target protein sequence as completely as possible. Then the glycosylation sites in the protein are analyzed. After determining the peptide segments with glycoside modifications, the target protein is re-digested to retain the glycosides on the peptide segments. During the LC-MS/MS analysis, a combination of ETDHCDCID fragmentation methods is used to obtain as much information as possible for secondary fragmentation of the peptide. The accurate information of glycosylation sites and the sugar type at these sites in the target protein is obtained by analyzing the secondary spectra of glycosylation modified peptides through software analysis and manual identification.
