Hyocholic acid (HCA) is a trihydroxy bile acid predominantly found in pig bile, recognized for its crucial role in bile acid metabolism and interactions with gut microbiota. Structurally, HCA is distinct from human bile acids, such as cholic acid and chenodeoxycholic acid, due to the presence of an α-hydroxy group at the 6th position. In comparison to muricholic acids in rodents, HCA also differs in the positioning and configuration of its hydroxyl groups. Within the liver, HCA undergoes conjugation with taurine or glycine to form taurohyocholate or glycohyocholate, which are subsequently secreted into the bile. In the intestine, bacterial 7α-dehydroxylation converts HCA into the secondary bile acid, hyodeoxycholic acid. These distinctive features make HCA particularly significant in the study of bile acid metabolism, liver function, and gut microbiota regulation.
