      iTRAQ-based quantitative proteomics is a technique that employs isobaric tagging reagents to label peptides from different protein samples. The iTRAQ reagents are chemically identical in mass but release reporter ions of distinct masses during tandem mass spectrometry (MS/MS). This allows for the simultaneous identification and quantification of proteins from multiple samples within a single liquid chromatography-mass spectrometry (LC-MS/MS) run, making it a powerful tool for comparative proteomic studies.

       

      iTRAQ consists of a peptide-reactive group, a balance group, and a reporter group. The reactive group covalently binds to peptides, ensuring consistent and uniform labeling. The balance group offsets the mass added by the reporter group, so all labeled peptides have identical overall masses. The reporter group enables protein quantification by indicating the relative abundance of each peptide from different samples. In the initial MS1 scan, these peptides appear as single peaks due to their equal masses. During MS2 analysis, fragmentation causes the balance group to be lost neutrally, while the reporter group is released as a reporter ion with a unique mass specific to each sample's tag. By measuring the intensities of these reporter ions, the relative abundance of peptides across samples can be accurately determined. This allows precise quantification of proteins, making iTRAQ-based proteomics a powerful tool for comparative protein analysis.
