    MALDI Mass Specrometry Analysis Service

The Matrix-Assisted Laser Desorption/Ionization (MALDI) mass spectrometry analysis service uses laser energy absorbed by a matrix to generate ions from macromolecules with minimal fragmentation, allowing for the measurement of the molecular weight of biomolecules. MALDI mass spectrometry primarily utilizes a MALDI ion source and a Time-of-Flight (TOF) mass analyzer, forming the MALDI-TOF mass spectrometer. The principle of the MALDI mass spectrometry analysis service involves spotting various microbial samples with an excess of matrix solution onto a sample plate. After the solvent evaporates, a co-crystal of the sample and matrix is formed. Laser energy is then used to irradiate the crystal. The matrix absorbs energy from the laser, which causes the sample to adsorb. A charge transfer occurs between the matrix and the sample, leading to the ionization of the sample molecules. The sample ions acquire the same kinetic energy in the accelerating electric field. After high-pressure acceleration and focusing, they enter the TOF mass spectrometer for mass analysis. The detector detects ions with different mass-to-charge ratios (m/z), and a pathogen protein fingerprint is generated by plotting m/z ratios on the x-axis and ion peaks on the y-axis. This fingerprint is then compared with a database for identification.Based on mass spectrometry technology combined with quantitative analysis and pathway analysis, MtoZ Biolabs offers MALDI mass spectrometry analysis service that can perform protein analysis, metabolomics analysis, lipidomics analysis, and a variety of other analyses.

      Case Study

      1. Deubiquitinase analysis for drug discovery based on high-throughput matrix-assisted laser desorption mass spectrometry (MALDI-TOF)

This article presents a mass spectrometry-based screening method for deubiquitinase (DUB) activity. Using MALDI mass spectrometry, the single ubiquitin signal generated by DUB cleavage of ubiquitin chains is detected to assess DUB enzyme activity. The MALDI mass spectrometry analysis service offers advantages such as high throughput, the use of physiological substrates, and the sensitivity and reliability of mass spectrometry readings. This service can be applied in drug discovery for screening efficient and specific DUB modulators.
