    N-Terminal Amino Acid Determination

      N-terminal amino acid determination is a crucial technique in protein chemistry and proteomics. This method focuses on identifying the initial amino acid at the N-terminus of a protein or polypeptide chain, playing a significant role in foundational research and application development. Although effective, its complex procedures and large sample requirements make it more suitable for shorter peptides or protein fragments. In contrast, mass spectrometry, paired with modern sample preparation techniques, offers a higher throughput solution with greater sensitivity and detailed analysis of complex samples.

       

      In mass spectrometry, proteins are typically subjected to enzymatic or chemical cleavage to reveal and release the N-terminal amino acid for detection. Techniques such as liquid chromatography-mass spectrometry (LC-MS) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) provide high sensitivity and resolution, delivering robust support for N-terminal amino acid determination.
