Peptidome is a discipline dedicated to studying endogenous peptides within biological systems or protein degradation products under specific conditions. Peptides typically refer to sequences smaller than 10 kDa. Current research focuses on endogenous peptides, low-molecular-weight proteins, bioactive peptides, disease-specific peptides, and protein degradation products. Peptidomic service investigates these peptide fragments to uncover the biological significance of cytokines, antimicrobial/antiviral peptides, flavor peptides, and disease biomarkers, playing a crucial role in medicine, food science, and other fields. Analysis Workflow
The fundamental approach for peptidomics and proteomics sequencing services is essentially the same. The experimental workflow for peptidomic service consists of the following steps: First, the sample undergoes preprocessing to extract peptides. Common extraction methods include size exclusion chromatography, ultrafiltration, and organic precipitation. Once the peptides are obtained, the sample is subjected to reduction and alkylation treatment to break disulfide bonds, facilitating subsequent peptide detection. Next, the purified peptides are introduced into a liquid chromatography-mass spectrometry (LC-MS) system for mass spectrometric analysis, generating spectra. These spectra are then searched against a spectral database using specialized software to obtain qualitative and quantitative information on the peptides. Finally, bioinformatics methods are applied to analyze various properties of the peptide samples, providing as much relevant information as possible to clients.

It is worth noting that in the entire peptidomic service experimental workflow, enzymatic digestion is generally not performed. Instead, the database search parameters are typically set to "unspecific" to ensure more accurate spectral library predictions. The peptidomic service provided by MtoZ Biolabs consider the research needs of many customers for small peptides. During the mass spectrometry acquisition process, we will relax the collection range to 100-1500m/z to ensure effective collection of small peptides. In the final deliverables, MtoZ Biolabs provides not only qualitative and quantitative peptide results but also multiple physicochemical properties of peptides, including peptide length, amino acid composition, isoelectric point, and hydrophobicity. Additionally, based on client requirements, we offer both de novo sequencing and database search strategies.
