      1. Single-cell Proteomics Differentiates Arabidopsis Root Cell Types

      SCP is an emerging approach to resolve cellular heterogeneity within complex tissues of multi-cellular organisms. The Arabidopsis root represents a well-characterized organ with defined cell types, making it an excellent system for benchmarking SCP in plants. We examined single cells isolated from the cortex and endodermis of Arabidopsis roots using SCP and identified 3763 proteins from 756 root cells, of which 596 proteins were enriched in either the cortex or endodermis and were able to distinguish between these closely related plant cell types. This study demonstrates that SCP can resolve neighboring cell types with distinct functions, thereby facilitating the identification of biomarkers and candidate proteins to enable functional genomics.
