Protein methacrylation is a newly identified type of lysine acylation modification, classified under post-translational modifications (PTMs). This modification occurs when a methacryloyl group is covalently bound to lysine residues via glutaryl transferase, influencing key biological processes such as gene expression, chromatin remodeling, protein-protein interactions, and signal transduction. Methacrylation shares isomeric similarity with lysine crotonylation (Kcr), as both exhibit an identical mass shift (+68.023 Da) in mass spectrometric analyses. However, their subtle structural variations confer distinct biological functions and regulatory mechanisms, making the ability to distinguish between these modifications critical for understanding their roles in disease and cellular processes.
