17α-Ethinylestradiol (EE2) is a synthetic estrogen whose structure resembles that of the natural estrogen estradiol. The chemical structure of EE2 includes an ethynyl group substituted at the 17α position, a modification that imparts significant biological activity and stability, thereby markedly reducing its metabolic rate in the body and prolonging its duration of action. Metabolomics analysis, utilizing high-throughput techniques such as mass spectrometry and nuclear magnetic resonance, can comprehensively elucidate the metabolic pathways and mechanisms of EE2 within the body.
Compared to traditional metabolic analysis methods, metabolomics can detect a wider variety of metabolites and concentration changes, providing more comprehensive information on the metabolic network. Through this approach, both primary and secondary metabolic pathways of EE2 can be identified, including biochemical reactions involving specific enzymes, which helps in understanding the metabolic differences of EE2 in different tissues and organs. Metabolomics analysis can identify various metabolites of EE2 and their structural characteristics, including active metabolites and potentially toxic ones. This is crucial for elucidating the pharmacological effects and toxicological properties of EE2. For instance, some metabolites may have greater biological activity or toxicity than the parent drug; metabolomics analysis can help discover these key metabolites and investigate their potential impacts.
