One approach involves hydrolyzing protein samples into individual amino acids, followed by derivatization for HPLC detection to obtain qualitative and quantitative results for each amino acid, forming an amino acid profile. The experimental workflow for HPLC-based amino acid profiles analysis service includes hydrolyzing protein samples in 6 N hydrochloric acid under sealed conditions for 22 hours. The hydrolyzed amino acids are derivatized using AccQ Tag reagents, which convert primary and secondary amino acids into stable fluorescent derivatives. Separation is performed using the Waters AccQ•Tag amino acid analysis column, and the quantification of 17 amino acids is achieved using a single-point quantification method.

      2. Amino Acid Profiles Analysis Service Based on Amino Acid Automatic Analyzer

      The use of an amino acid automatic analyzer to detect hydrolyzed single amino acids eliminates the need for derivatization with reagents. It directly separates amino acids through liquid chromatography, and the quantification of 17 amino acids is achieved using the single-point quantification method. This approach provides accurate and reliable results while offering a convenient and efficient detection method.

      3. Amino Acid Profiles Analysis Service Based on LC-MS Technology

      The amino acid profiles analysis service based on liquid chromatography-mass spectrometry (LC-MS) follows a workflow similar to HPLC. Amino acids are derivatized using AccQ Tag reagents. The Waters ACQUITY UPLC I-CLASS system is employed for chromatographic separation, while the Waters XEVO TQ-S Micro tandem quadrupole mass spectrometer performs mass spectrometric analysis. Quantification is achieved using the MassLynx software to calculate peak areas, applying the standard curve method. This approach enables both qualitative and quantitative analysis of more than thirty amino acids and their derivatives with high precision.
