    Analysis of Sialic Acid Content in Recombinant Protein Vaccines

      Recombinant protein vaccines are a class of vaccines that do not contain complete pathogens and are prepared from specific protein antigens produced by heterologous expression systems. Glycosylation is an important post-translational modification of recombinant protein vaccines. Sialic acid (SA) is a derivative of neuraminic acid, an acidic amino sugar with nine carbon atoms and a pyranose structure. It is usually located at the non-reducing end of the glycoprotein glycoside, among which N-acetylneuraminic acid (NANA) is the main form of sialic acid. The content of sialic acid affects the half-life and biological activity of the recombinant protein vaccine. Therefore, the analysis of sialic acid content has become an important step in the development of biopharmaceuticals.

       

      Currently, the determination of sialic acid content in recombinant protein vaccines mainly includes resorcinol method, enzyme method, and high-performance liquid chromatography (HPLC). The resorcinol method is a traditional method for measuring sialic acid content. This method is fast, accurate and has good reproducibility, but it has poor sensitivity and a narrow linear range. Although the enzyme method is accurate and has high sensitivity, it also has high cost, more complex experimental operations, and a narrow linear range. In comparison, HPLC has many advantages such as fast speed, accuracy, high sensitivity, good reproducibility, and wide linear range when determining the content of sialic acid, which greatly makes up for the shortcomings of the first two methods. Therefore, HPLC is widely used in the determination of sialic acid content in bioproducts such as recombinant protein vaccines.
