    Determination of Antibody Drug Extinction Coefficient

      Antibody drugs are a class of drugs that treat diseases through artificially synthesized antibodies, binding specifically to target molecules to achieve therapeutic purposes. Common types of antibody drugs include monoclonal antibodies, artificially synthesized antibody fragments, immunotoxins, antibody-drug conjugates, etc. The development and production of antibody drugs not only require an understanding of the biological characteristics of proteins but also need to ensure their safety and effectiveness through various experimental techniques. Among them, accurately measuring the concentration of antibody drugs is a crucial step, which involves the determination of the extinction coefficient.

       

      The extinction coefficient, also known as the molar absorbance coefficient or molar extinction coefficient, is a parameter widely used in spectroscopy and molecular physics. The extinction coefficient describes the ability of a substance to absorb light of a specific wavelength. The determination of the extinction coefficient is usually carried out by an ultraviolet-visible spectrophotometer, which can measure the absorption of a sample to light of a specific wavelength and then obtain the extinction coefficient. If the molar extinction coefficient is known, it can be used to determine the concentration of protein in the solution. A common method for determining the extinction coefficient is the spectrophotometric method, which is also the method most bioproducts currently use to determine the extinction coefficient. In the determination of the extinction coefficient of antibody drugs, the absorbance at 280nm of the aromatic amino acids of the protein (such as tyrosine, phenylalanine, and histidine) is usually selected because these amino acids have strong absorption to light of this wavelength.
