    Disulfide Bond Localization in Antibody Drugs

      Antibody drugs are a type of protein drugs produced by genetic engineering technology, mainly including monoclonal antibodies and antibody-drug conjugates, etc. They have a high degree of specificity and can accurately target and neutralize disease-related molecules. Among these antibody drugs, disulfide bonds are crucial for maintaining the three-dimensional structure and functional stability of the antibodies.

       

      In the research and production process of biological drugs, it is essential to accurately detect and control the disulfide bonds in the antibodies. Incorrect disulfide bond pairing may lead to drug failure, or even potentially cause safety risks. The methods for disulfide bond localization mainly include mass spectrometry, nuclear magnetic resonance, X-ray, and auxiliary chemical methods.

       

      Mass spectrometry is a commonly used method for disulfide bond localization, with high sensitivity and resolution. By cutting the antibody protein into multiple peptide fragments with specific enzymes or chemical reagents, then using mass spectrometry to analyze these fragments, the mass of protein fragments can be directly measured, thus determining the position of disulfide bonds. In addition, mass spectrometry can handle relatively large sample volumes and has relatively low requirements for sample purity.
