    Endogenous Fluorescence Detection of Recombinant Protein Vaccines

      Recombinant protein vaccines are vaccines prepared by expressing and purifying the antigen proteins of pathogens in engineered cells (such as bacteria, yeast, mammalian cells, and insect cells) using genetic engineering technology. Compared with other types of vaccines, recombinant protein vaccines are more costly but have significantly better safety profiles. The intrinsic fluorescence of a protein mainly originates from the aromatic amino acids in its structure, such as tryptophan, tyrosine, and phenylalanine, which emit fluorescence under ultraviolet light. When the protein structure changes, such as folding, unfolding, or binding to ligands, its intrinsic fluorescence changes accordingly. Therefore, by measuring the intrinsic fluorescence of recombinant protein vaccines, it is possible to obtain information about their structure and monitor possible denaturation during the preparation and storage processes, thus assessing the stability of recombinant protein vaccines.

       

      The detection of the intrinsic fluorescence of recombinant protein vaccines is typically carried out using a fluorescence spectrometer, which can accurately measure the intensity and wavelength of the protein fluorescence to obtain the fluorescence spectrum. By analyzing the fluorescence spectrum, reaserchers can obtain much important information about recombinant protein vaccines. Although intrinsic fluorescence detection is a powerful tool, it also has some limitations, such as potential interference from quenchers or signal overlap when the sample contains large amounts of aromatic amino acids. Therefore, intrinsic fluorescence detection typically needs to be combined with other analytical methods (such as circular dichroism and nuclear magnetic resonance) to obtain more comprehensive and accurate information.
