    Lactylation Proteomics Analysis

      "Lactylation Proteomics Analysis" refers to the systematic study of protein lactylation modification. Lactylation is a post-translational modification (PTM) in which a protein reacts with lactate on a certain lysine residue to form a modification. In recent years, with the rapid development in the fields of metabolomics and proteomics, it has been found that lactate is not only an intermediate product of metabolism, but also plays an important role in various levels such as cell signal transduction and gene expression regulation, and lactylation is one of the mechanisms.

       

      Protein lactylation modification may affect the function, location, interaction, and stability of proteins. Therefore, systematically analyzing protein lactylation modifications can help us understand the relationship between cell metabolic status and protein function.

       

      Analysis Workflow

      1. Sample Preparation

      Proteins are extracted from the biological sample and digested, usually using trypsin to break down the proteins into peptides.

       

      2. Enrichment of Lactylated Modification Peptides

      Since the abundance of modified peptides in the protein digest is low, specific affinity chromatography methods are used to enrich peptides with lactylation modifications.

       

      3. Mass Spectrometry Analysis

      The enriched peptides are separated and identified by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Mass spectrometry can provide precise mass and fragment ion information of peptides, thereby determining the lactylation site.

       

      4. Data Analysis

      Mass spectrometry data are analyzed by bioinformatics tools to identify lactylated modified peptides and proteins and to quantify their abundance.

       

      5. Biological Function Study

      After determining the lactylated proteins and modification sites, researchers usually need to perform further biological experiments to explore the impact of these modifications on protein functions.

       

      Lactylation proteomics analysis is a constantly developing field and is of great significance for understanding the link between cell metabolism and signal transduction, and the pathophysiological process of related diseases.
