N-Acetylserotonin (NAS), also known as Normelatonin, is an intermediate in the biosynthesis of melatonin from tryptophan. It is synthesized from serotonin by the enzyme arylalkylamine N-acetyltransferase (AANAT) and is converted to melatonin by the enzyme acetylserotonin O-methyltransferase (ASMT). NAS is mainly found in the pineal gland, retina, and gastrointestinal tract. In addition to its role in the production of melatonin, NAS has been shown to have potent antioxidant, anti-inflammatory, and neuroprotective properties.
N-Acetylserotonin plays a crucial role in the regulation of circadian rhythms and sleep-wake cycles. Irregularities in its production or metabolism can lead to sleep disorders, mood disorders, and various neurological conditions. Furthermore, understanding the biosynthesis and regulation of N-Acetylserotonin can aid in the development of new therapeutics for these conditions.
