SDS-PAGE serves as a conventional method for purifying oligonucleotides by size. However, higher-resolution chromatography is required when oligonucleotides are similar size or contain modifications that hinder separation. The preferred method for oligonucleotide purification is ion-pair reversed-phase high-performance liquid chromatography (IP-RP-HPLC), which can be coupled with MS for detailed oligonucleotide mass characterization. At MtoZ Biolabs, we utilize a HPLC platform to develop and validate methods for analyzing oligonucleotide purity.
