    Protein Fingerprinting Service

Protein fingerprinting, also known as peptide mass fingerprinting (PMF), is a widely-used mass spectrometry technique for protein identification. First developed independently by several research groups in 1993, this method involves enzymatically cleaving proteins into peptides and measuring their mass-to-charge ratios (m/z) for rapid, precise identification. Unlike whole-genome sequencing, Protein fingerprinting does not require complete protein sequence data. Instead, it identifies proteins based solely on the peptide mass data compared to theoretical data in established databases.

MtoZ Biolabs offers highly efficient and accurate Protein Fingerprinting Service for the rapid identification and quantification of proteins. Our service is extensively applied in basic research, biomarker discovery, proteomics, and quality control for biopharmaceuticals, providing robust data support for both research and industrial applications.

      Analysis Workflow

      1. Sample Separation

Protein samples are first separated using 1D or 2D PAGE.

      2. Digestion

Selected bands or spots are excised, and the proteins are enzymatically digested into peptides.

      3. Mass Spectrometry Analysis

Peptide mass-to-charge ratios are measured using mass spectrometers such as MALDI-TOF or LC-MS/MS.

      4. Data Comparison

The resulting mass spectrometry data are matched against theoretical peptide masses in a database to identify the proteins.

      5. Report Generation

A comprehensive report detailing the protein identification results is provided.

      Sample Submission Suggestions

Samples that can be tested by Protein Fingerprinting Service include:

      • Low-complexity samples: Gel spots or bands, approximately 1-2 cm in length.
      • Medium-complexity samples: Protein mixtures, including column-purified samples.
      • Moderate-to-high complexity samples: Protein mixtures such as IP-derived samples, secreted proteomes, or subcellular organelles (e.g., mitochondria, endoplasmic reticulum, lysosomes).
      • High-complexity samples: Whole cell lysates, tissues.

      FAQ

Q1: Can Protein Fingerprinting Service handle complex mixtures?
This service is best suited for single proteins or simple mixtures. For samples containing multiple proteins, we recommend MS/MS analysis to achieve high accuracy and resolution. This approach allows precise identification even in complex samples. For higher-throughput needs, our LC-MS/MS service is also available as an ideal option.

Q2: If my sample has already been separated by 1D/2D PAGE, is further liquid chromatography necessary?
For samples with clear PAGE bands, we typically excise, digest, and proceed with mass spectrometry. However, for complex samples or bands with multiple protein components, our nano-LC system provides additional separation, enhancing resolution and identification accuracy. MtoZ Biolabs will select the optimal approach based on your sample to ensure reliable results.
