      Analysis Workflow

      MtoZ Biolabs' protein full-length sequencing service comprises three primary steps:

       

      1. Protein Digestion

      In the initial step, proteins are enzymatically digested into smaller peptide fragments. Traditional sequencing methods often rely on trypsin digestion, which typically results in about 60% coverage. To enhance sequence coverage, MtoZ Biolabs incorporates up to six protease combinations, ensuring more comprehensive detection of peptide fragments.

       

      2. LC-MS/MS Analysis

      Following peptide separation using liquid chromatography (LC), tandem mass spectrometry (MS/MS) is employed to analyze the mass-to-charge ratios of the peptides, allowing for precise identification of their sequences.

       

      3. Data Analysis

      Using advanced algorithms, the MS/MS data is processed to reconstruct the full protein sequence while identifying post-translational modifications (PTMs) and other chemical modifications.
