Protein Identification Services
Protein identification is a technique used to detect proteins in samples through experimental and data analysis methods. Currently, protein identification services are primarily based on mass spectrometry analysis, including protein sequence analysis, post-translational modification (PTM) studies, molecular weight determination, and N/C-terminal sequencing. These services help determine the types, quantities, and functions of proteins in a sample, providing researchers with critical data support. For instance, in common molecular biology experiments like co-immunoprecipitation (CO-IP), the conventional approach involves performing a CO-IP experiment, consulting literature to identify potential interacting proteins, purchasing expensive antibodies for Western blot (WB) validation, and potentially obtaining successful results—though more often resulting in false positives, which can be time-consuming and labor-intensive. Mass spectrometry-based protein identification services effectively address this issue. A portion of the CO-IP sample can be subjected to mass spectrometry for identification, followed by data analysis to identify proteins potentially interacting with the target protein based on metabolic pathways or interaction networks. This narrowed-down list can then undergo WB validation.

The protein identification services provided by MtoZ Biolabs are not limited to CO-IP samples but also applicable to a wide range of sample types, including tissue samples, protein solution samples, gel bands, and processed beads. Upon receiving client samples, our laboratory first extracts and purifies the proteins, performs SDS-PAGE electrophoresis, and conducts quality control on the protein solutions. Portions of the protein solution are then subjected to in-solution digestion, or specific bands of interest are excised from the gel for in-gel digestion. The proteins are digested into peptides using proteases (commonly trypsin), and the resulting peptides are desalted and purified. These peptides are then analyzed using liquid chromatography-mass spectrometry (LC-MS) to acquire data. Specialized database search software is used to match the mass spectrometry data with species-specific databases, yielding detailed protein information. A comprehensive bioinformatics analysis follows to help clients identify the content of interest in their samples.
