Protein Sequencing Service by Edman Degradation Analysis
MtoZ Biolabs utilizes the SHIMADZU PPSQ-33A analyzer to provide protein sequencing service by Edman degradation. Samples include but are not limited to purified protein products, antibodies, and protein vaccines, etc. Using our sequencing system, the sequence information of the 30 amino acids at the N-terminus can be determined. Using a specific protein loading system, the 60-70 amino acids at the N-terminus can be determined.
Service Advantages
1. High Sensitivity Detection
MtoZ Biolabs uses the SHIMADZU PPSQ-33A analyzer, which integrates an advanced HPLC system and UV-VIS SPD-20A detector. This setup is capable of detecting PTH-AA at low picomole level, ensuring high precision and sensitivity in detection.
2. Broad Applicability
Protein sequencing service by Edman degradation can analyze up to 30 amino acid residues and is particularly effective for confirming the N-terminal sequences of antibodies, vaccines, peptide therapeutics, protein drugs, recombinant protein, ect.
3. High-Resolution Separation
The advanced HPLC system can accurately distinguish amino acids, i.e. I/L, and Q/K, allowing efficient analysis of complex samples, including proteins with post-translational modifications or specific domains.
Sample Submission Requirements
1. Electroblotted Samples
Proteins separated by SDS-PAGE should be transferred to PVDF membranes. It is recommended to stain the membranes with Coomassie Brilliant Blue or Ponceau S, avoiding silver staining. Thorough washing of the PVDF membranes is required to remove potential contamination from glycine buffer.
2. Liquid Samples
Samples should be prepared at a concentration of 1-10 µg with a purity greater than 90%. Buffers should exclude Tris, glycine, SDS, and other potential contaminants to ensure accurate sequencing.
