Purity and Size Analysis of Inactivated Vaccine Virus Particles

Inactivated vaccines refer to vaccines made by cultivating and inactivating pathogenic microorganisms and are a common type of vaccine. For inactivated virus vaccines, the structural protein of the virus is one of the most important components of the vaccine, which includes surface protein and capsid protein. The structural protein of the virus is a key factor in the virus life cycle and virus infection of the host, and is also the main target of the immune response induced by the vaccine.

Viral particle purity refers to the ratio of active ingredients in the vaccine to other components (such as proteins, nucleic acids, cell residues, and inactivating agents). High-purity viral particles can provide a stronger immune response and reduce the risk of adverse reactions. Therefore, evaluating and controlling the purity of viral particles is an important part of the vaccine production process. Electrophoresis and high-performance liquid chromatography techniques are commonly used to analyze purity.

The size of the viral particles can affect the immunogenicity of the viral particles, thereby affecting the efficacy of the vaccine. The size of the viral particles directly relates to the interaction between the virus and host cells, including the efficiency of cells taking up the virus and the type of immune response induced by the virus. Accurate measurement of the size of viral particles can help us better understand the biological properties of viruses, optimize vaccine design, and evaluate the quality of vaccines. Dynamic light scattering (DLS) is one of the most commonly used methods to determine the size and radius distribution of particles or polymers in solution samples, and is also widely used to measure the size of viral particles. DLS measures the diffusion coefficient of particles in the medium being measured through the autocorrelation function (ACF) of photons, and can also calculate the size distribution of spheres and describe in detail the movement of particles in the medium being measured.
