MALDI-TOF analysis of peptide mixtures is capable of tolerating moderate amounts of buffers and salts, with peptide fragments predominantly producing singly charged ions, making MALDI-TOF MS an ideal tool for peptide mass mapping. To confirm the theoretical sequence of a protein/polypeptide sample using MALDI-TOF, enzymatic digestion is first performed on the sample, followed by the verification of the peptide mass fingerprinting (PMF) via MALDI MS. Subsequent MS/MS analysis is used to validate spectral peaks, which are then compared against your provided theoretical sequence to generate a peptide coverage mapping. Advantages and Limitations of Service
1. Advantages
This method is a classic approach to protein identification, characterized by its simplicity, speed, and broad application prior to the emergence of tandem mass spectrometry.
2. Limitations
Peptides of similar mass increase the difficulty of matching and cannot achieve the identification of mixed proteins. It is not suitable for protein identification in species with incomplete databases, cannot analyze post-translational modification sites, and is not easily connected with liquid chromatography.
