    What Causes Baseline Drift During HPLC Sample Injection?

      To determine the amount of a 100-unit enzyme required for protein digestion, follow these steps:

       

      1. Determine the Total Amount of Protein

      For example, if you have 1 milligram (mg) of protein.

       

      2. Calculate the Required Enzyme Amount

      For Asp-N: If the recommended ratio is 50:1, the required amount of Asp-N is 1 mg / 50 = 0.02 mg.

       

      For Glu-C: If the recommended ratio is 20:1, the required amount of Glu-C is 1 mg / 20 = 0.05 mg.

       

      3. Convert Enzyme Amount to Units

      Determine how many units are present per milligram of enzyme. This information is typically provided in the enzyme's product manual or specifications.

       

      Assume each milligram of Asp-N or Glu-C contains X units.

       

      4. Calculate the Required Number of Units

      For Asp-N: 0.02 mg × X units/mg

       

      For Glu-C: 0.05 mg × X units/mg

       

      5. Determine the Actual Amount to Use

      If 100 units of enzyme correspond to Y milligrams, use the ratio of the required units to Y to calculate the actual amount of enzyme to add.

       

      Throughout this process, consult the enzyme's product manual for precise unit content and enzyme activity data to ensure accurate calculation of the required enzyme amount.
