Two-Dimensional Gel Electrophoresis Service
Two-dimensional gel electrophoresis (2-DE) is a powerful and widely used method. It combines IEF and SDS-PAGE. First, proteins are separated by their isoelectric point (pI) in IEF, and then by SDS-PAGE based on molecular weight. Each protein spot on the two-dimensional gel can be eluted and identified by high-throughput mass spectrometry. Therefore, 2-DE is particularly suitable for comparing protein profiles between different tissues, different conditions, or between control and experimental samples.
Analysis Workflow
MtoZ Biolabs provides 2-DE services. The 2-DE service can separate thousands of proteins from complex mixtures for subsequent quantitative analysis.
1. Sample Preparation
The preparation process depends on the sample type.
2. First Separation by IEF
Load the protein mixture onto a pH gradient gel, and proteins migrate under the influence of an electric field.
3. Second Separation by SDS-PAGE
This method generally includes three steps: preparing the gel, equilibrating the pH gradient strips fixed in SDS buffer, and conducting electrophoresis.
4. Staining
Proteins are stained using various staining methods, such as silver staining and Coomassie Brilliant Blue staining.
5. Result Analysis
Besides protein separation, protein molecular weight, pI, and content can also be determined.
Sample Submission Requirements
We accept various types of samples, including but not limited to the following types:
1. Purified Protein
2. Frozen Cell Pellets or Tissue
3. Cells (such as Blood Cells) or Tissue
4. Serum/Plasma/Urine
5. Cell Cultures
6. Fermentation Cultures
7. Microorganisms
8. Plants
9. Others
Service Advantages
1. High Throughput
Analyze thousands of proteins with just one 2-DE.
2. High Resolution
Proteins are resolved by pI and molecular weight, and protein characterization is achieved through post-translational modifications that affect their charge state.
3. Advanced Bioinformatics Software
We have SameSpots, Delta2D, ImageMaster, and other software for detecting and quantifying protein spots.
4. Low Cost
2-DE is relatively cheaper compared to mass spectrometry analysis.
5. Flexibility
MtoZ Biolabs provides customizable analytical services and designs optimal experimental plans to achieve your research goals within your budget.
Applications
1. Protein Profiling
2. Protein Isoform Distribution Analysis
3. Protein Quantification
