    Edman Based Protein Sequencing Service

      The Edman degradation method, proposed by Pehr Edman in 1950, is a protein sequencing technique that determines the amino acid sequence of a protein by sequentially removing and identifying the N-terminal amino acids of a polypeptide chain. The Edman degradation method is significant in protein research. It provides precise determination of the N-terminal amino acid sequence of proteins, particularly suitable for short peptides and purified proteins. It helps verify the translation start sites of proteins, ensuring accurate sequence information. By understanding the N-terminal sequence of proteins, it is possible to infer functional domains and active sites.

      Analysis Workflow

      1. Sample Preparation

      Purifying the target protein to ensure high purity and appropriate concentration.

       

      2. Coupling Reaction

      Under alkaline conditions, the N-terminal amino acid of the protein reacts with PITC to form a phenylthiocarbamyl derivative.

       

      3. Cleavage Reaction

      The coupled product is cleaved under acidic conditions to produce a PTH amino acid.

       

      4. Identification and Analysis

      HPLC is used to separate and identify the PTH amino acid, determining its structure.

       

      5. Cycle Repetition

      Repeating the above steps for the remaining polypeptide to sequentially analyze the entire sequence.
