Protein Analysis: Can Protein WB Expression Be Verified by qPCR
Western Blot (WB) expression of proteins and qPCR (quantitative polymerase chain reaction) actually analyze different levels of biological information. WB is mainly used to detect protein expression levels, while qPCR is used to detect mRNA expression levels of genes. Although there is some correlation between them, they measure different biomolecules.
Western Blot (WB) is an immunoassay technique used for qualitative and quantitative analysis of target protein expression levels in cells or tissues. It involves protein electrophoresis, transfer to a membrane, detection of the target protein with specific antibodies, signal detection, and analysis.
qPCR is a method for quantitatively detecting the amount of specific mRNA molecules. It uses fluorescently labeled probes or primers to monitor the fluorescence signal during the polymerase chain reaction process in real time. By measuring the mRNA levels of a specific gene, we can understand its transcription activity.
If you wish to verify changes in protein expression levels, it is better to use another protein detection method, such as ELISA or mass spectrometry. However, if you want to understand the transcriptional regulatory mechanisms behind protein expression changes, using qPCR to detect the mRNA expression of the corresponding gene makes sense. Note that changes in mRNA levels do not necessarily directly lead to changes in protein levels, as translation regulation, protein stability, and degradation are other influencing factors.
