    What Are the Differences Between AKTA Fast Protein Liquid Chromatography and HPLC in Protein Purification?

      Both AKTA and HPLC are chromatographic techniques commonly used for the purification of proteins and other biomacromolecules. However, they differ significantly in terms of design, applications, and operational characteristics:

       

      Primary Applications

      1. AKTA

      The AKTA system, developed by GE Healthcare (now Cytiva), is specifically designed for the purification of biomolecules, particularly proteins. It is widely used in affinity chromatography, ion-exchange chromatography, and size-exclusion chromatography.

       

      2. HPLC

      High-performance liquid chromatography (HPLC) is designed for the separation, identification, and quantification of various types of molecules, including small molecules, peptides, nucleic acids, and proteins.

       

      Operating Pressure

      1. AKTA

      Operates at relatively low pressures, typically below 500 psi.

       

      2. HPLC

      Can function at pressures up to several thousand psi, particularly in ultra-high-performance liquid chromatography (UHPLC).

       

      Column Characteristics

      1. AKTA

      Uses columns with larger-diameter particles and larger pore sizes, which are well-suited for biomacromolecule separation.

       

      2. HPLC

      Employs columns packed with smaller-diameter particles and smaller pores, enhancing resolution but requiring higher operational pressure.

      Typically equipped with UV detectors and, in some cases, conductivity or other specialized detectors.

       

      2. HPLC

      Supports a variety of detectors, including UV, fluorescence, refractive index, and mass spectrometry.

      Offers a high degree of automation and user-friendly operation, making it particularly advantageous for large-scale protein purification.

       

      2. HPLC

      While automation is available, it often requires more manual intervention, especially for complex sample separations.

       

      Sample Volume Handling

      1. AKTA

      Typically used for biological samples in the microgram-to-gram range.

       

      2. HPLC

      Depending on the system and column size, it can process sample volumes from nanograms to milligrams or larger.

       

      Resolution and Separation Efficiency

      1. AKTA

      Provides sufficient resolution for biomacromolecule separation.

       

      2. HPLC

      Due to its use of smaller-particle columns, it generally achieves higher resolution and better separation efficiency.

       

      While both AKTA and HPLC are effective for protein purification, they serve different purposes based on experimental needs. The AKTA system is advantageous for large-scale, automated protein purification but requires a significant investment and technical expertise. HPLC, on the other hand, is widely used for high-resolution separations across a broad range of sample types but typically involves longer separation times and greater manual involvement. The choice between these methods should be based on experimental objectives, required purity levels, sample throughput, and available laboratory resources.
