What Are the Methods for Obtaining Protein Sequences?
Protein sequences can be obtained through two primary approaches:
Gene Sequencing and Translation
Protein sequences can be inferred from genetic information using DNA sequencing technologies. First, the sequence of a protein-coding gene is determined through DNA sequencing. Then, bioinformatics tools translate the coding regions (exons) into the corresponding amino acid sequence based on the genetic code.
Protein Mass Spectrometry Analysis
This method involves direct analysis of protein samples. First, enzymes such as trypsin cleave proteins into smaller peptides. These peptides are then analyzed using mass spectrometry to determine their mass and sequence characteristics. Finally, bioinformatics tools reconstruct the full protein sequence by assembling peptide sequence data.
Each approach has distinct advantages. Gene sequencing and translation are effective for predicting protein sequences from known genes, while mass spectrometry is particularly useful for obtaining sequence information directly from protein samples, especially in the study of protein modifications and variants.
